Quantification of HBsAg in serum: characteristics of the assays
نویسنده
چکیده
Abstract Introduction The presence of hepatitis B virus (HBV) surface antigen (HBsAg) in serum is the hallmark of HBV infection, and qualitative tests for its detection have been used since 1969. Recently, HBsAg quantification has been applied for monitoring chronic HBV infection during its natural history as well as for the prediction of the response to treatment. As HBsAg titres are becoming an integral part in the monitoring of chronic hepatitis B, knowledge of the characteristics of HBsAg quantitative assays is of great importance. Two automated assays are currently commercially available, namely the Architect QT (Abbott Laboratories) and the Elecsys HBsAg II Quant (Roche Diagnostics). Although the correlation between the currently approved methods for HBsAg quantification is quite good, the use of the same assay in the monitoring of individual patients with chronic HBV infection appears to be mandatory. The aim of this review is to discuss the characteristics and comparisons of available HBsAg quantitative assays. Conclusion Correlation of HBsAg quantification is good, but using the same assay in the monitoring of patients with chronic HBV infection is mandatory. Dilution steps for commercial assays minimize human error. We call for further studies to be conducted so that we are able to improve the techniques we have and further our understanding.

Is Quantitative HBsAg Measurement a Reliable Substitute for HBV DNA Quantitation?
Background: Hepatitis B surface antigen (HBsAg) is one of the main proteins of HBV envelop and its serum quantitative measurement is the most common quantitative test for monitoring the progress of Chronic Hepatitis B. Although measurement of serum HBV DNA copy number is a gold standard method for displaying viral load, the test is relatively expensive and it is not readily available everywhere...

The coding sequence for the hepatitis B virus surface antigen (HBsAg) was used as a new reporter gene for studies on eukaryotic promoter activity and upstream regulatory sequences. The data observed in transfection assays were comparable to results obtained with conventional chloramphenicol acetyltransferase (CAT) assays, as was demonstrated using various transcriptional regulation sequences. T...

Objective Dot Blot (DB) assay provides highly specific results, but usually not reliable for quantification of antibody production. The need for a more objective DB assay to provide a better definition of the immune status, against HIV antigens, promoted this study to be done to develop a quantitative DB assay. Material and Methods Dot blot (DB) strips for antibodies directed to human immuno...
متن کاملHepatitis B Surface Antigen Quantification across Different Phases of Chronic Hepatitis B Virus Infection Using an Immunoradiometric Assay
BACKGROUND/AIMS Quantification of hepatitis B surface antigen (HBsAg) is an emerging serologic test and may be useful for identifying treatment strategies for chronic hepatitis B (CHB). This study aimed to evaluate HBsAg titers during the natural course of CHB and identify correlations between HBsAg titers and hepatitis B virus (HBV) DNA concentrations across different CHB phases measured using...
